چکیده

Particles of certain rat tissue preparations contain a phosphate ester hydrolase that destroys carbamyl phosphate. This activity is similar to that of the alkaline phosphatases. Both have the same molar activities, pH curves, and particulate localization. Both are activated when solubilized with 1butanol and are inhibited by adenine nucleotides and inorganic phosphate. Their reactions are not additive in the same assay. They have the same distributions in rat tissues and tumors, with highest activities in adult kidney and in fetal and adult small intestine. The two activities also increase in parallel during development from fetal to adult kidney. Aspartate transcarbamylase activity in homogenates containing the hydrolase can be measured qualitatively by inhibition of the hydrolase or by raising the carbamyl phosphate concentration to allow for its hydrolysis during the assay.

P-178: Separation and Identification of Alkaline Phosphatase Isozymes during Pregnancy

Background: Alkaline phosphatase (ALP), (EC 3.1.3.1) is a hydrolase enzyme responsible for removing phosphate groups from various molecules in the body. In humans, ALPis present in all tissues such as liver, bile duct, kidney, bone, and the placenta which detection of its activity is so useful in molecular biology. Pregnancy is associated with normal physiological changes that assist fetal surv...

Evaluation of the organophosphorus hydrolase enzyme activity in creams and investigation of its stability

The main purpose of this project is investigation of the organophosphorus hydrolase (OPH) enzyme activity in water in oil (w/o) and oil in water (o/w) creams and investigation of the OPH enzyme stability in formulated creams. OPH enzyme was extracted and purified from strain flavobacterium. The w/o and o/w creams were prepared using different formulations. In order to achieve an emulsion with m...

Inhibitors of neutral cholesteryl ester hydrolase.

p-Nitrophenyl N-butyl, N-octyl, and N-dodecyl carbamates and a newly synthesized diethyl phosphate compound were studied as potential inhibitors of the cholesteryl ester hydrolases of Fu5AH rat hepatoma cells. Whole homogenates of Fu5AH cells were used as an enzyme source for the assay of cholesteryl ester hydrolase activity. All four compounds led to marked inhibition (70-80%) of neutral chole...

Purification and characteristics of aspartate transcarbamylase from Pseudomonas fluorescens.

Aspartate transcarbamylase from a Pseudomonas fluerescens has been purified to homogeneity. The enzyme has a molecular weight of 360,000. It is composed of 2 apparently identical subunits with molecular weights of 180,000. Activity can be recovered from the enzyme denatured with mercaptoethanol and sodium dodecyl sulfate when the dodecyl sulfate is dialyzed away. The renatured enzyme has the mo...

Urethan carcinogenesis and nucleic acid metabolism: in vitro interactions with enzymes.

In the light of suggestions in the literature linking the car cinogenic action of urethan (ethyl carbamate) with the inhibi tion of an enzymatic step in nucleic acid metabolism (par ticularly pyrimidine synthesis), a series of enzymes has been assayed in vitro at concentrations of urethan which are supralethal in mice. These enzymes were: ornithine transcarbamylase; the enzymes leading to the s...
